Orthologous series in chromosomes of homoeologous group 4. Vrn-Am2 was located in T. monococcum (Dubcovsky et al., 1998) on chromosome 5Am on the 4Am translocated region. Vrn-Am2 was mapped to the distally located Xwg114-5A - Xwec87-5A region (Shindo et al., 2002).
Vrn-2 is a zinc-finger/CCT domain transcription factor (ZCCT1) (Yan, Loukoianov et al., 2004), and repressor of flowering down-regulated by vernalization and short days (Dubcovsky et al., 2006). Reduction of Vrn-2 transcripts in transgenic hexaploid winter wheat accelerates flowering (Yan, Loukoianov et al., 2004).
The Vrn-2 locus produces two linked and related proteins designated ZCCT1 and ZCCT2, characterized by the presence of a putative zinc finger and a CCT domain. Deletions and mutations involving both the ZCCT1 and ZCCT2 genes are frequent in diploid wheat and are associated with recessive alleles for spring growth habit (Yan, Loukoianov et al., 2004).
Among the cultivated tetraploid and hexaploid wheat species the Vrn-B2 gene is generally functional whereas the Vrn-A2 gene is not. At least one functional copy of Vrn-2 combined with homozygous recessive alleles at all three Vrn-1 loci is required to confer winter growth habit in hexaploid wheat (Distelfeld et al., 2009).
A triple Vrn2 mutant (PI 676269; synthetic vrn2-null) is available in hexaploid wheat combining the non-functional vrn-A2 allele present in most polyploid wheats with a Vrn-B2 deletion from tetraploid wheat, and a non-functional vrn-D2 allele from Ae. tauschii (Kippes et al., 2016).
Vrn-H2(sh/sgh1) occurs in barley chromosome 4H (Takahashi and Ysuda, 1971) and is probably orthologous to Vrn-Am2 based on comparative maps (Dubcovsky et al., 1998; Laurie et al., 1995).
