- Assembly and loading of an electro-elution cuvette. (a) Insert a collection cup into the small aperture of an assembly joint. (b) Add 1X TAE to the cup/joint unit until a reverse meniscus is visible at the top of the joint. (c) Place a Miracloth square onto the reverse meniscus. (d) Insert the frosted end of a glass tube into the large aperture of the assembly joint. Push the tube in as far as it will go. The Miracloth should be stretched across the bottom of the glass tube. (e) The cuvette is now completely assembled and ready for loading. (f) Fill the cuvette with sterile 1X TAE buffer. Drop an agarose cube containing size-selected DNA into the cuvette. The cube should drift through the buffer until it rests on the Miracloth stretched across the bottom of the glass tube. Use a glass rod to gently push the cube to the bottom of the tube if it does not drift down by itself.
