Abstract
Whole-plant regeneration by somatic embryogenesis was studied in wheat cultivars Inqilab-91 and Pavon-76. Embryogenic callus was obtained by culturing the seeds of both cultivars on gel-solidified Linsmaier and Skoog's medium with 3.5 mg 2,4-D/litre. The callus cultures were maintained by subsequent subcultures onto fresh media at 4- to 5-week intervals. The embryogenic calluses cleared of any surrounding non-embryogenic callus were grown on gel-solidified basic Murashige and Skoog's medium with 3% (w/v) sucrose after at least 2 subcultures. Seven combinations of BAP (6-benzylaminopurine [benzyladenine]) and IAA (0 + 0.1, 0.5 + 0.1, 2.5 + 0.1, 5.0 + 0.1, 0.5 + 0, 2.5 + 0 and 5.0 + 0 mg/litre) and one control were assayed. The highest frequency of green spot formation and plant regeneration (84% in Inqilab-91 and 52% in Pavon-76) were obtained with 0.5 + 0.1 mg BAP + IAA/litre. Higher concentrations of BAP in the media was toxic and fatal for calluses in some cases. Significant variation in callus initiation and in vitro regeneration was observed between the cultivars
