Abstract
A study was conducted to evaluate the utility of ISSR markers as a tool for the identification of inbred lines and for the construction of genetic map of rye. Sixteen rye inbred lines and a mapping population composed of 94 individual F SUB 2 plants from the 544 x Ot0-20 intercross were used. Twelve primers, which generated strong and reproducible amplification products, were selected out of 57 primers tested. The selected primers revealed 29 polymorphic bands which allowed the identification of lines and the estimation of their genetic diversity. A dendrogram developed on the basis of genetic similarities proved to be generally consistent with the pedigree data. Out of eight markers used for the analysis of F SUB 2 intercross population, six showed segregation consistent with 3:1 expectation for a single locus. The presented results of ISSR marker segregation suggest that this PCR-based method should be suitable for the construction of rye genetic map. However, its efficiency seems to be comparable with that of RAPD
